Multifunctional Activity Of Resveratrol And Gallic Acid Co-Loaded In Viscous Structured Vesicles

Resveratrol and gallic acid, naturally occurring phenols, were co-loaded in phospholipid vesicles intended for protecting the skin from external injuries, such as oxidative stress and microbial infections. The phenolco-loaded liposomes were prepared using biocompatible phospholipids dispersed in water. To improve vesicle stability and applicability, the liposomeswere modified with glyceroland/or poloxamer407.As displayed by cryogenic transmission electron microscopy and photon correlation spectroscopy, the vesicles were spherical, unilamellar and small in size: liposomes and glycerosomes were around 70 nm in diameter.The presence of glycerol and poloxamerincreased the viscosity of the vesicle systems, which positively affected the stability of the dispersions.

The low cytotoxicity and great abilityof the vesicles to protect fibroblasts from chemically-induced oxidative damage was demonstrated. Additionally, an improvement of the in vitro antimicrobial activity of the phenols, when loaded in vesicles, was confirmed against different skin pathogens, such as Candida spp.,P. aeruginosa, S. aureus, and Streptococcus spp. Further, the superior ability of the vesicles to promote the accumulation of both phenols in the skin was demonstrated, as well as their therapeutic potential againstoxidative inflammation caused by TPA (phorbol ester)on mouse skin, asindicated by a significant reduction of both oedema and leukocyte infiltration.
